Use of dihydro-resveratrol in the treatment of pancreatic fibrosis in chronic pancreatitis

Abstract

Chronic pancreatitis is a long-standing inflammation of pancreas due to pancreatic fibrosis. Patients with chronic pancreatitis will suffer from abdominal pain, diarrhea, nausea and weight loss as a result of fibrotic replacement of parenchyma. Long term pancreatic exocrine damage will lead to impairment of endocrine function which gradually cause type 3c diabetes.

Dihydro-resveratrol is a stilbene that can be naturally found in Dendrobium spp., Dioscorea spp., Bulbophyllum triste and wine. Besides, the compound has also been identified as microbial metabolite of resveratrol derived from the elimination of the central carbon-carbon double bond by hydrogenation of gut microbiota. Previous report suggested that dihydro-resveratrol may serve as therapeutic agent for the management of acute pancreatitis as its strong anti-inflammatory and antioxidant bioactivity. In this study, by utilizing different means of biochemical and physiological approaches, we found that dihydro-resveratrol can ameliorates the activation of pancreatic stellate cells (PSCs) and the production of extracellular matrices upon the stimulation of different pro-inflammatory cytokines. In addition, we found that dihydro-resveratrol possess protective effect against cytokines induced damage to NIT-1 beta cells. Therefore, we suggest that the natural stilbene can be a potential remedial agent for treatment of pancreatic fibrosis in chronic pancreatitis.
