Abstract

目的:研究白藜芦醇(RSV)对活化的小鼠T淋巴细胞分泌IL 2和CD2 5的影响,并探讨其免疫抑制机制。方法:无菌分离小鼠淋巴结细胞,与不同浓度的白藜芦醇共同孵育1小时后,用多克隆刺激剂佛波醇酯和离子霉素刺激T细胞活化,继续培养6小时后收获细胞,进行胞内细胞因子染色,流式细胞仪检测IL- 2的分泌情况,RT- PCR检测IL -2mRNA的表达;2 4小时后检测CD2 5表达。结果:RSV对IL 2的分泌具有抑制作用,并呈剂量依赖性,同时RSV也能抑制T细胞表面活化分子CD2 5。结论:RSV对活化T细胞分泌的细胞因子IL- 2及IL- 2α链CD2 5的表达可能是RSV对T细胞具有免疫抑制作用的机制之一,并可能与T细胞活化通路的PKC NF -κB信号传导途径相关。

Objective: To study on the effects of resveratrol on IL-2 and CD25 expression in murine T activated lymphocytes. Methods: The lymphocytes were isolated and pre-treated with resveratrol for 1 h prior to activation with PDB plus ionomycin. After 6 hours stimulation,IL-2 secretion was determined by intracellular cytokine staining by flow cytometry. IL-2 mRNA expression was detected by RT-PCR. CD25 expression is determined by multi-color flow cytometric analysis after 24 hours stimulation. Results: RSV can inhibit IL-2 expression at mRNA and protein level. It also inhibit CD25 expression. Conclusion: The immunosuppressive mechanism of RSV maybe relative to the inhibitory effects on IL-2 mRNA and protein expression and CD25 expression, which may be relative to the T cell activated signal transduction of PKC-NF-κB pathway.
